Why is Dihexa studied?
Angiotensin IV is short-lived in biological matrices, which limits its usefulness as an experimental probe. The stabilised analogues in that series were prepared specifically to give investigators a longer-lived chemical tool for interrogating the same receptor pharmacology, and they were characterised alongside the parent peptide for that reason.1
How is Dihexa studied in the lab?
In the reported work the analogues were assessed in rodent models of scopolamine-induced cognitive deficit and in accompanying binding and activity assays; potency was compared across the series to establish structure–activity relationships for the stabilised backbone.1
What has preclinical research reported?
Angiotensin IV analogue chemistry
McCoy and co-workers reported that terminal modification of the angiotensin IV scaffold produced analogues that retained activity in the rodent cognition models used, while resisting the peptidase cleavage that inactivates the parent peptide.1
