DSIP entered the literature in 1977, when Schoenenberger and Monnier isolated and characterised a nonapeptide from cerebral venous blood and named it for the delta-EEG activity they were tracking.3 It is studied today mainly as a historically important, structurally simple neuropeptide: nine residues, no disulfide bridge, and an unusually well-documented isolation record.4

How is DSIP studied in the lab?

No single high-affinity DSIP receptor has been cloned, so mechanistic work is done indirectly — radiolabelled-peptide binding to brain membrane preparations, stability and transport measurements, and immunoreactivity mapping across tissues. The 1984 Neuroscience & Biobehavioral Reviews survey collected that body of work and remains the reference point for what is and is not established about the peptide.4

The originating PNAS report described the isolation and chemical characterisation of the delta-EEG-inducing nonapeptide from cerebral venous blood.3

Review of the preclinical record

Graf and Kastin’s 1984 review compiled the DSIP literature to that date, including binding, distribution and stability findings in animal preparations.4

Forms studied and handling

The material is supplied as a lyophilized (freeze-dried) powder. Freeze-drying is used for peptides because removing water suppresses the hydrolysis and aggregation routes that dominate degradation in solution, and the resulting amorphous solid is far more stable on storage than the corresponding liquid.5 Reported degradation routes for peptides in the solid state include deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and physical aggregation, all of which are accelerated by residual moisture, elevated temperature and repeated freeze-thaw cycling.6 Sealed vials are therefore held at −20 °C, protected from light, and reconstituted material is kept cold and used within a short working window. This causes the water in the peptide vial to sublimate directly from solid to gas, leaving behind a stable, crystalline white structure known as lyophilized peptide. The puffy white powder can be stored at room temperature until it is reconstituted with bacteriostatic water.

Storing vials on arrival

Once peptides have been received, it is imperative that they are kept cold and away from light. If the peptides will be used in the next several days, weeks or months, short-term refrigeration under 4°C (39°F) is generally acceptable.
