Thymosin Beta 4 (TB-500): Stimulating Hair Follicle Growth Through Stem Cell Activation and Angiogenesis

Activating Hair Follicle Stem Cells
Thymosin β4 is a naturally occurring 4.9 kDa peptide first discovered in the thymus and now recognized as a powerful regulator of wound repair, angiogenesis, and cellular migration. Within the skin, it is expressed in keratinocytes that originate from the bulge region of the follicle, a known reservoir of hair follicle stem cells.
Studies show that Thymosin β4 boosts migration of these clonogenic keratinocytes, enhances differentiation, and stimulates production of matrix metalloproteinase-2 (MMP-2), an enzyme critical for remodeling the extracellular matrix. These actions accelerate the transition of follicles into the anagen (growth) phase, leading to faster and denser hair regrowth.
Evidence From Animal Models: Enhanced Hair Cycling
While investigating wound healing in rats, researchers observed unexpected increases in hair growth at wound edges following topical application of Thymosin β4. Controlled studies confirmed that treated skin displayed twice as many anagen-phase follicles compared to untreated controls.
This effect persisted with continued treatment, but regressed within two weeks of stopping, demonstrating that Thymosin β4 directly influences follicular cycling. In shaved C57BL6 mice, topical application likewise triggered rapid regrowth and follicle activation verified by histological examination.
Angiogenesis Near Hair Follicles
Hair follicle activity is tightly linked to vascular supply. Thymosin β4 increases expression of VEGF and MMP-2 through the Wnt/β-catenin/Lef-1 signaling pathway, enhancing angiogenesis around follicles. By creating new microvasculature, Thymosin β4 ensures a robust nutrient and oxygen supply that supports active hair growth.
Role in Hair Follicle Development and Regeneration
Thymosin β4 also participates in developmental regulation of follicles. It activates signaling pathways such as MAPK, PKC, and Wnt to increase VEGF production, which in turn stimulates endothelial cell proliferation and hair follicle development.
In genetic mouse models, animals over expressing Thymosin β4 (KTP mice) displayed faster and thicker hair regrowth compared to wild-type (WT) mice, while knockout (KO) mice lacking the peptide had severely delayed regrowth.
Accelerating Hair Growth After Shaving
Although the onset of hair development after birth was unchanged, the speed of regrowth after shaving was significantly increased in Thymosin β4–over expressing mice compared to controls. This suggests that while Thymosin β4 may not alter initial follicle formation, it plays a key role in follicle reactivation and regrowth cycles.
