ARA-290 Calms Astrocyte and Microglial Inflammation

Research indicates that ARA290 can reduce spinal inflammatory mediators such as CCL2, Iba-1, GFAP, and NMDA receptor subunits (NR1, NR2A, NR2B) following nerve injury. This suggests that ARA290 modulates inflammatory responses in both microglia and astrocytes. Experimental and clinical evidence supports the notion that ARA290 reprograms a pro-inflammatory environment into one that promotes healing and tissue repair.

ARA-290 Protects Transplanted Islets and Inhibits Macrophage Activation

ARA290 has been shown to suppress intrahepatic inflammatory responses following pancreatic islet transplantation (PITx). It protects transplanted islets from cytokine-induced apoptosis and reduces secretion of pro-inflammatory cytokines such as IL-6, IL-12, and TNF-α. In treated animals, blood glucose levels improved significantly compared to controls. Additionally, ARA290 lowered expression of monocyte chemoattractant protein-1, macrophage inflammatory protein-1β, IL-1β, and IL-6 in liver tissue, highlighting its anti-inflammatory and protective effects.

Microglial Modulation by ARA-290

ARA290 effectively calms microglial activity in the spinal cord. In animal studies, doses of 10 and 30 μg/kg reduced microglia reactivity after nerve injury compared to untreated controls. Astrocyte activity remained unchanged, but suppression of microglial responses correlated with decreased neuropathic pain symptoms. This suggests a direct mechanistic link between central inflammation suppression by ARA290 and relief from neuropathic pain.

Neuroprotection in Diabetic Retinopathy

ARA290-derived licensed peptides such as pHBSP regulate microglial activation in diabetic retinas. While overall microglia numbers remain stable, pHBSP promotes dendritic morphology and reduces amoeboid microglia. Additionally, pHBSP restores anti-inflammatory IL-10 levels and normalizes elevated pro-inflammatory cytokines TNF-α and IL-6. This results in a healthier retinal environment and protects against vascular and neural damage associated with diabetes.

Macrophage Clearance and Polarization

ARA290 enhances the phagocytic activity of macrophages, promoting the clearance of dying cells instead of inducing inflammation. Classically activated M1 macrophages are pro-inflammatory, while alternatively activated M2 macrophages support tissue repair. ARA290 helps shift the balance toward M2 polarization, improving the M1/M2 ratio during kidney injury and reducing tissue damage.

Immune Modulation and Autoimmune Protection

ARA290 suppresses lymphocyte proliferation both in vitro and in vivo. In experimental autoimmune neuritis (EAN) rats, ARA290 reduced CD4+ Th cell numbers and spleen mononuclear cell proliferation. These results suggest a protective effect against systemic autoimmune diseases like lupus.

Anti-Inflammatory and Metabolic Effects

ARA290 and derivatives such as pHBSP promote M2 macrophage polarization, reduce M1 pro-inflammatory activity, and decrease susceptibility to diet-induced insulin resistance in mice. Treatment also protects retinal capillaries in diabetic models, preventing degeneration and preserving tissue integrity.
